Summary:Within the brain, imidazoline‐2 binding sites (I2BS) exhibit specific expression in reactive astrocytes and are implicated in various pathophysiological processes, such as analgesia, inflammation, Alzheimer's disease, Parkinson's disease, and glial tumors. In article number 20230037, Wanqing Li, Yuqing Qiao, and their co‐workers present an overview of the I2BS PET ligands by focusing on their chemotypes and PET imaging outcomes. Of note, [11C]BU99008 was a milestone for I2BS PET imaging and has been used in clinical PET imaging of both Alzheimer's disease and Parkinson's disease. Furthermore, the advances of I2BS drug discoveries were also discussed, thereby providing new clues to the development of more potent PET ligands targeting I2BS for clinical use.
