Summary: | DTH reactions in the skin represent an increasing cause of inflammatory dermatoses typically exemplified by allergic contact dermatitis (eczema). The "oxazolone mouse ear swelling assay" has been developed as a simple acute model for contact allergy in the skin. In a search for new topical anti-inflammatory drugs acting against eczema, our first objective was to characterize the animal model used for our screening purposes in terms of: kinetics of the vascular and cellular components of the inflammatory reaction; pharmacological modulation by known anti-inflammatory agents. We were also interested to see whether the injection of oxazolone into sub-cutaneous air-pouches in sensitized mice could provide an easy method for the study of cellular migration phenomena in this model. |
