Summary: | As an effective traditional Chinese medicine, pinosylvin displays anti-cancer effects on multiple cancers, but its role in esophageal squamous cell carcinoma was obscure. Thus, the aim of this paper was to explore whether pinosylvin could exert anti-cancer activity in esophageal squamous cell carcinoma, as well as its potential mechanism. Esophageal squamous cell carcinoma cells (ECA109 and TE1) were exposed to pinosylvin. The cell viability, invasion, and migration were assayed by Cell Counting Kit-8, transwell, and wound-healing assays, respectively. Corresponding gene and protein expressions were assessed with the employment of western blot and quantitative real-time PCR. Gain-of-function experiments were conducted for the determination of the involvement of syntaxin-6 in esophageal squamous cell carcinoma. The capabilities of esophageal squamous cell carcinoma cells to invade and migrate were reduced after pinosylvin treatment. Moreover, pinosylvin reduced the expression levels of syntaxin-6 and integrin α3 but enhanced vasodilator-stimulated phosphoprotein expression. Upon syntaxin-6 overexpression, the decreased integrin α3 and increased vasodilator-stimulated phosphoprotein expressions caused by pinosylvin in ECA109 and TE1 cells were reversed. In addition, syntaxin-6 overexpression weakened the inhibitory effects of pinosylvin on esophageal squamous cell carcinoma cell invasion and migration. In conclusion, we demonstrated the pivotal role of pinosylvin in treating esophageal squamous cell carcinoma, and uncovered that it was via the syntaxin-6/integrin α3/vasodilator-stimulated phosphoprotein pathway that pinosylvin exerted anti-cancer activity in esophageal squamous cell carcinoma cells.
