Summary:A technique for process analysis was developed based on flow-injection analysis (FIA) combined with diode-array spectrophotometry. The method demonstrates the advantages of coupling single-line FIA with diode-array detection, utilizing the stopped-flow and double injection techniques with a dual-wavelength monitoring approach. A simple, robust FIA hardware system was designed and rigorously tested for on-line process application. A fed-batch fermentation served as the test bed for the development of this system, monitoring selected analytes of interest, viz., glucose, ammonium ion and total protein.
