Summary:The effects of actinomycin D (AMD) at different concentrations and periods of treatment on RNA synthesis have been investigated in meristematic cells of Allium cepa L. roots. The analysis was accomplished by electropheresis on SDS agarose gels of nucleic acid extracts from 3H-uridine-labelled cells. The rate of uptake of the tritiated antibiotic was also estimated. The results show that AMD impaired ribosomal RNA synthesis in plant cells. As the concentration increased and treatment became longer, the inhibition of synthesis was greater. The delay in the inhibition caused by AMD was not due to a deficient uptake of the antibiotic.
