Summary:In our previous study, intracerebroventricular administration of thromboxane A2 (TxA2) receptor blockers and its systhesis inhibitors attenuated the nitric oxide-induced elevation of plasma catecholmines in rats. Based on these results, we have suggested that TxA2 is probably involved in central activation of adrenomedullary outflow (Neuroscience, 1998). Then, in the present study, effects of TxA2 mimetics on plasma levels of adrenaline (Ad) and noradrenaline (NA) were examined in urethane-anesthetized rats. U-46619 (100ng/animal) and I-BOP (2.5, 5.0ng/animal) microinjected into the paraventricular nucleus of hypothalamus (PVN) dose-dependently elevated plasma levels of Ad and NA (Ad>NA). These agents applied into other regions adjacent to the PVN were without effect. The present results confirm our suggestion that TxA2 in the PVN is probably involved in central activation of adrenomedullary outflow in rats.
