Summary: | We previously reported that diabetic rats 8 weeks after streptozotocin(STZ) treatments showed the tendency of hypercoagulability despite vulnerability of liver cells (J. Atheroscler. Thromb. 4, 29-35, 1997). However, studies on each mRNA level of coagulation factors have not yet been determined. In the present study, fibrinogen (Aα chain and Bβ chain) and prothrombin mRNA levels of liver homogenate were investigated by RT-PCR analysis. After 50 mg/kg of 517 i.v. injection (Wistar rats 6 weeks), VLDL-LDL-lipids and GPT level were increased, and activities of plasma coagulation factors (fibrinogen, ATIII, thrombin-ATIII complex) tended to increase at 72 hrs. There were no significant changes in fibrinogen and prothrombin mRNA levels at 24, 48 and 72 hrs except mild decrease 1 week after STZ injection. These results suggest that acute hyperglycemic rats induced by STZ caused mild changes in plasma concentration of coagulation factors and hepatocyte capacity of synthesizing coagulation proteins. The relation between plasma concentration of coagulation factors and their mRNA levels determined by RT-PCR, in STZ-induced diabetic rats, will be discussed. |
