Summary:We previously reported that ibudilast inhibits platelet aggregation and relaxes basilar artery by potentiating PGI_2 action. In this study, we investigated the influence of ibudilast on PGE_1 -induced inhibition of superoxide anion(O^- _2 ) generation in guinea pig peritoneal neutrophils stimulated by fMLP(10^-7 M) . PGE_1 (10^-7 M, 10^-6 M) inhibited O^- _2 generation and caused the elevation of intracellar cAMP levels 4.5- to 14.3-fold. Ibudilast(10^-7 M, 10^-6 M) enhanced these changes by PGE_1 . Dibutyryl-cAMP inhibited O^- _2 generation and IP_3 formation by phospholipase C in response of receptor activation in 「 ^^3 H」inositol labeled neutrophils. These results suggest that cAMP plays an important role in the mechanism by which ibudilast inhibits O^- _2 generation in neutrophils.
