Summary:Many calcium antagonists are extensively used for therapy in patients with angina pectoris. We have reported that non-dihydropyridine calcium antagonists have some beneficial effects on the experimental model of ischemic myocardium, but dihydropyridine calcium antagonists do not. In the present study, therefore, we studied the effect of a new dihydropyridine calcium antagonist, methyl 2,6-dimethyl-4-(2-nitrophenyl)-5-(2-oxo-1,3,2-dioxaphosphorinan-2-yl)-1,4-dihydropyridine 3-carboxylate, on ischemic myocardium in anesthetized open-chest dogs. In order to make the heart ischemic, the left anterior descending coronary artery was occluded either incompletely or completely. Ischemia decreased myocardial pH and myocardial high-energy levels, and stimulated tissue anaerobic carbohydrate metabolism. Changes in myocardial energy and carbohydrate metabolism induced by ischemia were not modified by the dihydropyridine calcium antagonist used in the present study (3,10,30 μg/kg, i.v.). This calcium antagonist did not reduce the ischemic influence on myocardium caused by coronary artery occlusion.
