Summary:The cardiomyopathic hamster (CMH), BIO 14.6 strain, is a well-known model of hypertrophic cardiomyopathy. To precisely evaluate the effect of Bay K 8644 on the heart of CMH, the following experiments were conducted by the use of Langendorff perfused preparations. We examined the relationship between the left ventricular developed pressure (LVP) and external Ca^2+ concentrations. In control hamsters, FIB strain, the LVP increased with increasing concentrations of Ca^2+ at 4, 8, 24, 40 weeks of age. However, the LVP of CMH was reduced and then insensitive to increase of Ca^2+ at 24 and 40 weeks of age. On the other hand, using SDS-PAGE we examined whether contractile proteins in CMH and F1B hearts changed with aging. There was no significant difference in the electrophoretic pattern between both strains up to 24 weeks of age. We will discuss the effect of Bay K 8644 on CMH hearts in relation to development of cardiomyopathy.
