Summary:Effects of KC-404, a novel cerebral vasodilator, on experimental amnesia were studied using a one-trial passive avoidance task in mice and were compared with those of nicardipine, idebenone, aniracetam and physostig-mine. The amnesia was elicited by electroconvulsive shock (ECS. 100 V, 0.5 sec) or 100% CO2 exposure (25 sec), performed immediately after training, and by injection of cycloheximide (CXM, 120 mg/kg, s.c.) at 30 min before training. The retention trial was carried out 24 hr after the acquisition trial. Drugs were administered after training or 30 min before the retention trial. KC-404 showed significant improving effects on the ECS-induced amnesia at 5-20 mg/kg, i.p., the CXM-induced amnesia at 20 mg/kg, i.p., and the CO2 -induced amnesia at 10 mg/kg, i.p.. Physostig-mine had significant effects on the ECS- and CO2 -induced amnesia. Nicardipine (5-10 mg/kg, i.p.) did not exert any improving effects. These results suggest that KC-404, as distinct from a cerebral vasodilator such as nicardipine, has an improving effect on the experimental amnesia in mice and may be a useful drug for the treatment of memory disorders.
