Summary:Pleurisy was induced by injection of 0.5 ml of 1% kaolin saline into the right pleural cavity of male SD rats. The plasma exudation, determined by the dye amounts exuded for 20 min into the pleural cavity, showed two peaks at 20 min and 3-5 hr after kaolin. The volume of pleural fluid increased gradually up to 5 hr. Captopril(3 mg/kg,i.p.) potentiated markedly the exudation at 20 min(350%) and to a lesser degree at 3 hr (14%) and 5 hr(73%). The early phase(20 min) of exudation was inhibited by bromelain(BRM, 10 mg/kg,i.v. ) , mepyramine (MEP, 2.5 mg/kg,i.v. ) +methysergide(MSG, 3 mg/kg,i.v.) and BRM + MEP + MSG by 50%, 50% and 85%, respectively. The later phase (3 hr) of the exudation was also inhibited by BRM and MEP + MSG by 30% and 20%, respectively. Indomethacin(10, 30 mg/kg,p.o.) and 480156-S(10, 30 mg/kg,p.o.) showed 30-50% inhibition of the later phase of exudation, but not the early phase. These results indicate that the plasma exudation in the kaolin pleurisy was induced by kinins, histamine and 5-HT in the early phaser followed by the later phase, being composed of prostaglandins and kinins.
