Summary: | The chemical investigation of the methanolic leaf and root extract of Coffea canephora Pierre ex A. Froehner (Rubiaceae) led to the isolation and structure elucidation of seventeen known compounds including three alkaloids: caffeine (1), theobromine (2) and paraxanthine (3); two diterpenes: tricalysiolide B (4) and 16,17-dihydroxykauran-18-oic acid (5); six triterpenoids: lupeol (6), 3β-acetylated oleanolic acid (7), ursolic acid (8), friedelin (9), polycarpol (10) and squalene (17); two phytosterols: stigmasterol (11) and stigmasterol-3-O-β-D-glucopyranoside (12); two phenolic compounds: 3,5-dihydroxy-7,8,2ʹ,3ʹ,4ʹ-pentamethoxyflavonol (13), 2,3,5-trimethoxyphenol (14) and two miscellaneous compounds: asperphenamate (15) and chlorophyll A (16). The structures of the isolates were established by extensive spectroscopic and spectrometric analyses (1D and 2D NMR, ESI-MS) and by comparison with previously reported data. Compounds 10 and 14 were reported for the first time from Rubiaceae family, whilst compounds 13 and 15 were reported for the first time from Coffea genus. In addition, some isolates were tested for their antibacterial and antifungal effects against various strains. Compounds 5 and 8 displayed moderate antibacterial activities against Shigella dysenteria with MIC value of 125 μg/mL, whilst compound 8 showed moderate antifungal activity against Candida krusei with MIC value of 125 μg/mL. Furthermore, the chemophenetic significance of the isolates is discussed in details.
