Summary:Homogeneous enzyme membrane immunoassays (EMIAs) have been developed for thyroxine (T 4 ) and human immuno-globulin G (IgG). Liposomes tagged with T 4 and containing alkaline phosphatase were used as a model system for optimization of (a) liposome membrane attack in the presence of T 4 antiserum and guinea pig complement and (b) subsequent measurement of unmasked enzyme activity. Using a one-incubation-step format, the concentration of T 4 in human serum samples was measured. Alkaline phosphatase activity was inversely proportional to the concentration of T 4 in the sample. The T 4 values obtained by EMIA correlated well with values obtained by radioimmunoassay (r = 0.96). Likewise, there was an excellent correlation between IgG values obtained by EMIA and radialimmuno-diffusion or nephelometry (r = 0.99).
