Summary:The capability of the Klenow fragment deficient in 3′→5′ exonuclease activity (KFexo − ) to incorporate 2′-deoxyisoinosine (disol) opposite canonical bases and 5-methyl-2-deoxyisocytidine (d 5Me isoC) was examined. The triphosphate of disol is incorporated opposite d 5Me isoC designed as its pairing partner. It is also incorporated opposite T and G according to a pairing scheme involving its minor tautomeric form (the O 2 H tautomer).
