Summary: | 1. Portions of stem pith from Cryptostegia grandiflora (Roxb.) R. Br., approximately 4 mg. in fresh weight, were grown on various media in culture tubes which were placed on an Auxophyton at room temperature in diffuse light. 2. Pith from different twigs possessed differing meristematic potential, which appeared to be inversely correlated with distance from the apical meristem. Rate of proliferation of pith from different plants also varied. 3. Copious proliferation of parenchymatous cells of pith was obtained on a medium containing modified Knop's solution, 2% sucrose, and 5-7.5% coconut milk by volume. Indoleacetic acid (IAA) was a relatively poor growth stimulator. 4. Either IAA or coconut milk is required for growth of excised pith of C. grandiflora, but explants of bark, which included all the bark tissues, proliferated without the addition of a growth factor. 5. No growth of the portions of latex tubes present in the pith explants was detected. |
