Summary: | The uptake, intracellular distribution, and radiotoxicity of several Auger electron emitters in cultured mammalian cells have been investigated, following an 18 h incubation of the cells with the various radionuclides. The mean lethal cellular uptakes (D37 values at 37% survival) of the radionuclides in mBq are: 1.5 (125IUdR), 2.6 (123IUdR), 4.8 (77BrUdR), 230 (51Cr), 145 (75Se), and 45 (201Tl). These radiotoxic indices are compared with the calculated localised energy deposition by the Auger electrons using theoretical estimates of the electron spectra for the radionuclides. The results emphasise the importance of intracellular and intranuclear localisation of the radionuclides as well as the quality of the Auger electrons in understanding the cytocidal effects of tissue-incorporated Auger emitters. Classical dose estimates are not adequate to account for the toxicity of these radionuclides. |
