Summary: | We have identified multiple Sry loci on the rat Y chromosome. In this study, we asked whether all Sry copies are expressed in adrenal gland, kidney and testis, and if each locus is expressed at the same level in each tissue. RNA from five 15 week‐old male rats was reverse transcribed, and cDNAs amplified using primer sets that produce amplicons of varying lengths that correspond to specific Sry copies. Fluorescent labeled PCR products were detected using the ABI 3130xl Genetic Analyzer and analyzed using GeneMapper v4.0 software (Applied Biosystems). Transcript amount from a particular Sry locus was calculated as a percentage of total Sry transcript present in a given sample. Results show that transcripts from each Sry locus are present in all three tissues and the relative amount of each copy expressed differs among tissues.
