Summary: | Toll like receptors (TLRs) play a pivotal role in sepsis, but their contribution to sepsis‐induced impairment of HPV is unknown. To assess the impact of TLRs on HPV, left lung pulmonary vascular resistance (LPVR) was measured before and during left mainstem bronchus occlusion (LMBO) in wild‐type (WT), TLR2−/−, and TLR4−/− mice. We studied HPV 22 hours after IV challenge with saline, or a TLR2 agonist (Pam3Cys, 20mg/kg), or a TLR4 agonist (lipopolysaccharide, LPS, 20mg/kg). In additional mice, PMN were depleted prior to the challenge with saline, Pam3Cys or LPS. Furthermore, circulating white blood cell (WBC) concentration and lung myeloperoxidase (MPO) activity were measured 22 hours after challenge with saline, Pam3Cys or LPS.
