Summary: | The method of solid-phase extraction (SPE) for the concentration and clean-up of tobacco extract samples during solanesol analysis was proposed in this work. A column (200 mm x 4 mm i.d.) packed with 0.10 g silica gel (with particle size of 70 μm, porosity of 0.5 and surface area of 400 m² g-¹) was used as SPE cartridge. Several extraction parameters, such as sample loading flow (0.3-7 mL min-¹), sample volume (5-50 mL), the column adsorption capacity and elution were evaluated to provide a fast, quantitative and reproducible SPE method. A mean solanesol recovery was 97.5 ± 1.6% (mean ± sd) and mean intra- and inter-day reproducibility was higher than 97%. It can be used in the analysis of solanesol in tobacco extracts. |
