Summary: | Previous in vivo studies demonstrated a stimulated hepatic capture of α-aminoisobutyric acid (AIB) following exposure of rats to whole-body γ-irradiation. This observation has been confirmed by the present in vitro studies using liver slices for the study of amino acid uptake. AIB accumulated inside the cell against a concentration gradient which required an endogenous source of energy. This transport mechanism was observed to be Na+ and K+-dependent and sensitive to ouabain. Protein synthesis was needed for the stimulated AIB transport. The radiation-induced stimulation of hepatic amino acid capture was also observed with a second model amino acid, cycloleucine. However, it still remains to be established if these observations are generally applicable to the hepatic uptake of natural amino acids. |
