Summary:Monoclonal antibodies specific for the sodium channel blockers saxitoxin (STX) and tetrodotoxin (TDT) have been generated and characterized as to their abilities to protect against toxin-induced in vitro toxicity and to induce anti-idiotypic immune responses. In a continued effort to enhance the binding affinities of the anti-toxin monoclonal antibodies, two novel approaches have been undertaken. The first one involves the design of a new toxin conjugate for use in immunization of BALB/c mice. The second approach involves the in vitro stimulation with free STX or TDT of mouse spleen cells hyper-immunized in vivo. Monoclonal anti-TDT and anti-STX antibodies have been generated that are protective against TDT and STX toxicity, respectively, on nerve functions. Rabbit anti-idiotypic antibodies specific for TD2C5 anti-TDT have been generated, purified and used in immunization of mice in order to test the potential use of anti-idiotype as a vaccine in inducing a systemic anti-TDT immune response.
