Summary:Introduction: Acetylcholinesterase enzyme, found in the cholinergic terminal, is a specific choline esterase which hydrolyzes predominantly choline esters (acetylcholine--ACh) present in high levels in the brain. These Cholinergic neurons in the brain are significantly affected by circulating levels of estrogen. Choline and DHA (docosahexaenoic acid) are the major components of synaptic membrane, additionally choline is also a precursor for the neurotransmitter acetylcholine. Dietary supplementation of fenugreek seeds that contains phytoestrogens may limit the risks and side effects or in combination with Choline and DHA may be an alternative strategy to treat menopause induced neurodegenerative diseases. Objective: To evaluate the efficacy of fenugreek seed extract (FSE) and Choline with DHA on brain acetylcholinesterase activity in ovariectomized rat model. Methodology: Female Wistar 9-10 months old rats were divided into groups (n=6/group): (1) Normal Control, (2) ovariectomy (OVX), (3) Sham OVX, (4) OVX + Fenugreek (FG--hydro-alcoholic extract), (5) OVX + Choline-DHA, (6) OVX + FG + Choline-DHA and (7) OVX + Estradiol. Groups 2, 4, 5, 6 and 7 were subjected for surgical ovariectomy. Dietary supplementation was initiated 15 days after ovariectomy for a period of 30 days.
