Summary: | We aimedtoexaminethepresenceofZn,atraceelement,inosteoarthritis(OA)cartilageandmeniscusfrom patientsundergoingtotalkneejointreplacementforprimaryOA.WemappedCa2+ and Zn2+ at themesoscopic scalebyX-rayfluorescencemicroanalysis(X-ray) todeterminethespatialdistributionofthe 2elementsincartilage,X-ray absorptionnearedgestructurespectroscopytoidentifytheZnspecies,and X-ray diffractiontodeterminethechemicalnatureofthecalcification.Fouriertransforminfraredspectroscopy wasusedtodeterminethechemicalcompositionofcartilageandmeniscus.Ca2+ showeda heterogeneousspatialdistributioncorrespondingtothecalcificationswithincartilage(ormeniscus)or attheirsurface.Atleast2Zn2+ species werepresent:thefirstmaycorrespondtoZnembeddedinprotein (differentZnmetalloproteinsareknowntopreventcalcificationinbiologicaltissues),andthesecond maybeassociatedwithaZntrapinoratthesurfaceofthecalcification.CalcificationpresentinOA cartilagemaysignificantlymodifythespatialdistributionofZn;partoftheZnmaybetrappedinthecalcification andmayaltertheassociatedbiologicalfunctionofZnmetalloproteins. |
