Summary:Diuretic effect of a newly synthesized compound, 7-chloro-1-(2-methyl)benzoyl-2,3-dihydro-4(1H) -quinolinone-4-oxime-O-sulfonate potassium salt(M17055) , was investigated. In conscious Splague-Dawley rats, oral administration of M17055 caused diuresis at doses ranging from 0.3 to 100 mg/kg in a dose-dependent manner. Though the slope of dose-response curve of H17055 was less steep than that of furosemide, maximul diuresis of M17055 was about the same as that of furosemide and the potency of the diuretic effect of N17055 was about 20 times higher than that of furosemide with regard to the ED_50 , values of these compounds. Dose-response relationship of H17055 on Ca^++ excretion showed a little difference from those on fluid, Na^+ and Cl^- excretions, whereas furosemide showed no such difference. In anesthetized dogs, intravenous injection of M17055 also caused diuresis in a dose-dependent manner, and the potency of the diuretic effect was about 20 times higher than that of furosemide. These results suggest that M17055 is a high-ceiling diuretic and its potency is higher than that of furosemide.
