Summary: | In traditional Chinese medicine, Rhubarb was used for the treatment of some psychotic disease. Recently, we demonstrated the Rhubarb water extract possessed the anti-methamphetamine(MAP) and anti-aggrssive effect in rats, and the most active comcponent was the origometric proan thocyanidine RG-tannin. Therefore, we further evaluate the anti-psychotic properties of RG-tannin in Wistar male rats weighing 180-200g. RG-tannin 2mg/kg i.p. decreased the spontaneous motor activity and MAP-induced hyperactivity. Furthermore, RG-tannin 5mg/kg i.p. inhibited the hyper activity induced by microinjection of MAP 40ug/ul into nucleus accumbens (NAc), while no effect by intra striatal injection of MAP. RG-tannin 1mg/kg i.p. also markedly suppressed the hyperemotionality such as attack and struggle response in olfactory bulbectomized(OB) rats without causing ataxia, muscle relaxation and catalepsy. From these results, the activity of RG-tannin was higher than any other extract from Rhubarb. RG-tannin, therefore, possessed the anti-psychotic action via blocking mesolimbic dopamine function, especially NAc Without extrapyramidal signs. |
