Summary:15-HPETE is a 15-lipoxygenase metabolite of arachidonic acid and has been shown to be lethally toxic to endothelial cells (1). Our previous finding of the presence of 15-lipoxygenase activity in cultured astrocytes (2) suggests the possibility that 15-HPETE produced locally by astrocytes may exert deleterious effects on the neuronal cells nearby. Effects of 15-HPETE on cultured neuronal cells prepared from hippocampi of 19-day-old rat embryos were examined. When neurons were exposed to 10^-5 M 15-HPETE, they appeared to be healthy at first. Three hours after exposure, however, disintegration of both cell bodies and neurites was apparent, which was preceded in many cases by swelling of the soma. Glutathione, in the reduced form, effectively prevented the neuronal injury by 15-HPETE.
