Summary: | In this study, we investigated the effects of antihistaminic drugs and steroid in various strains of mice with experimental allergic edema induced by repetitive sensitization and challenge with ovalbumin using the method of Kohno et al. ICR, ddY and Balb/c mice were repeatedly sensitized by i.p. injection of 1 μg OVA/2mg Al(OH)_3 / 100 μl saline/animal. Mice were challenged by i.d. injection with 10 μg/site into the hind paw 10 days after the respective sensitizations. On the 3-5th challenges, edema was dose-dependently and reproducibly provoked, and biphasic reaction with peak responses at 0.5 and 4 hr after challenge was observed. On the 3rd challenge, mice were assigned taking the degree of hind paw swelling into consideration. On the 4th challenge, we studied the effect of intrapenitoneal administration and topical application of drugs on the biphasic edema. In all strtains, with intraperitoneal administration, diphenhydramine (DH) suppressed only the immediate phase edema, while dexamethazone (DEX) suppressed only the late phase edema. On the other hand, topical application of DH ointment inhibited immediate phase edema in all strains, while DEX ointment did not suppress late phase edema in ICR and Balb/c mice. These findings indicate that only for ddY strain mice did both intraperitoneal administration and topical application of DH inhibit the immediate phase response and DEX suppress the late phase response. This model is useful for the analysis on subchronic allergic reaction. |
