Summary: | We have recently reported that phenytoin and nifedipine-induced gingival proliferation is mediated by angiotensin II (Ang II) and possibly AT1 receptors for Ang II in cultured gingival fibroblasts derived from guinea pig and rabbit, and suggested that fibroblasts may possess Ang II receptors. In the present study, we characterized the Ang II receptor subtypes present in rabbit cultured gingival fibroblasts. Initial Western blot analysis identified the presence of AT1 and AT2 receptors in the fibroblasts. Subsequent radioligand binding assay showed that Ang II completely displaced [3H]Ang II binding, and that CV-11974 and PD123,319 maximally displaced up to approximately 60% and 40% of the total binding. These findings confirm that gingival fibroblasts contain both AT* and AT, receptor subtypes for Ang II. |
