Summary: | SUN N8075 is a novel compound which has dual blocking actions on Na+ and T-type Ca2+ channels with antioxidant properties. In this study, we examined the effects of SUN N8075 on cerebral infarction in rat pMCAO model. The pMCAO was conducted according to the modified method of Koizumi using 4-0 nylon suture, and the cerebral infarction volume was determined 24 hrs after MCAO by using TTC staining method. SUN N8075 was intravenously infused at the loading dose of 3 mg/kg/S min followed by maintenance dose of 0. 75 mg/kg/hr until histological evaluation, starting immediately, or 1, 2, 3, 4 hrs after MCAO. SUN N8075 (started immediately after MCAO) markedly reduced the cerebral infarction (34.1 ± 1.2 % vs. 25.8 ± 2.7 %, p < 0.05), and even 3 hrs delayed infusion still significantly reduced the cerebral damages. There were no significant differences in physiological parameters between two groups. These results indicate that SUN N8075 is a potent neuroprotective drug with an extended TTW. |
