Summary:In the course of study of some molecular properties of the ciliated protozoan Tetrahymena pyriformis was a function of its mode of growth, conventional thin sections were examined in the electron microscope. As the cells progressed in their growth from the ultradian or fast exponential mode to the infrathan or slow exponential (quasi-stationary) mode, their ultrastructural appearance was that of cells becoming progressively emptied of their normal endoplasmic constituents . The major portion of the endoplasm had no conspicuously evident contents other than the macronucleus . Suspecting the synthesis and storage of glycogen not only because of the dramatic gross rise in assays of anthrone-positive material in our own' and previous work, but also because of the abundant supply of D-glucose in the medium, we applied an ultracytochemical technique for the demonstration of glycogen particles, the periodic acid/ thiosemicarbazide/silver protein method of Thiery, to the sections.
