Summary:Measurements were made of the volume changes exhibited by isolated guard cell protoplasts (GCPs) of Commelina communis L. when exposed to a range of concentrations of external osmotica. In short-term incubation, GCPs behaved as osmometers and showed rapid volume changes in response to changing external osmotic pressure (Πo). In long-term incubation, GCPs prepared and incubated with added external KCl showed further slow changes in volume, in a manner suggesting that regulation of volume occurred. Protoplasts prepared and incubated without added external KCl had smaller volumes for a given value of Πo, and their ability to regulate volume in long-term incubation was reduced or absent. Treatment with fusicoccin caused an increase in both the volume and the K+ content of GCPs. The increase in volume continuing after the increase in K+ content had ceased, in a manner similar to that observed in walled guard cells in epidermal strips.
