[...]we evaluated the effects of Fc[epsilon]RI stimulation on CB myeloid DCs (mDCs). [...]anti-Fc[epsilon]RI stimulation of purified CB mDCs demonstrated only a modest decrease in LPS-induced production of proinflammatory cytokines (Fig 1, G), suggesting that the suppressive effects are enhanced when Fc[epsilon]RI activation occurs within a mixed population of cells. Because Fc[epsilon]RI stimulation had a greater impact on DC responsiveness in a mixed population of cells, we reasoned that a soluble factor produced by another Fc[epsilon]RI-expressing cell type, such as basophils, might contribute to the inhibitory effect. [...]mixed CB leukocytes were treated with LPS and/or physiologic doses of histamine and analyzed to quantify intracellular TNF-α production by CB mDCs. [...]our observation that Fc[epsilon]RI stimulation diminishes CB mDC responsiveness to LPS supports a growing body of evidence that cross-talk between IgE/Fc[epsilon]RI and TLR signaling pathways serves a regulatory purpose.2,6,7 Importantly, we reveal a novel pathway for the regulation of DC-driven innate immune responses in the newborn that can be mediated by Fc[epsilon]RI cross-linking and secretory products such as histamine.
