Summary:Intracellular cGMP levels were measured by enzyme inked immunosorbent assay (ELISA): The effect of thrombin, ANP and NPR-A or NPR-C expression on endothelial barrier function was assessed by 3 way ANOVA. Intracellular cGMP levels were similar in all 3 cells under basal conditions (1.24 ± 1.31 versus 0.44 ± 0.71 versus 0.51 ± 0.67, pmol/ml for NPR-A, NPR-C, Zs-green, respectively) and increased markedly in response to ANP, with an increase that was approximately 4-fold greater in cells that overexpressed NPR-A as compared to those that overexpressed Zs-green or NPR-C (523.4 ± 466.9 versus 166.2 ± 124.2 versus 136.6 ± 118.8 pmol/ml).
