Summary: | Rhynchostylis retusa (L.) Blume is a monopodial epiphytic orchid species with attractive flowers arranged in racemose inflorescence. Mass propagation of wild type orchids were generally done by regenerated seeds from the same fruit. The study aimed to know the genetic similarity level of R. retusa plants which are regenerated from the same fruit seeds produced by self-pollination. The seeds were sown on New Phaleonopsis (NP) in vitro culture medium supplemented by 1.5% coconut water (CO). Genetic similarity levels were analyzed using PCR based Random Amplification of DNA Polimorfism (PCR-RAPD) with OPA 15 and OPA 03 primers. The analysis was performed 19 months after sowing the seed (m.a.s). DNA were isolated from 10 plants using a modified Castillo method. The electroforegram of the PCR result was converted to binary data and analyzed using NTSYSpc software version 2.02. The DNA analysis shows that all plants which were tested have a high similarity of DNA character. |
