[...]the present study focused on the optimization of operational parameters of ultrasound aided extraction of zeaxanthin like extraction temperature, extraction time and solid (algal biomass) to liquid (solvent) ratio from marine green microalga Dunaliella tertiolecta (NIOT 141) using response surface methodology (RSM). The algal biomass used in this study was cultured in De Walne's medium22 prepared in natural sea water (salinity 34.23 psu and pH 8.01) obtained from Kovalam coast, Tamil Nadu, India under laboratory conditions with a light intensity of 140pmol photon m2 s" 1, with 14:10 light/dark regimes at a temperature of 25 ± 1°C with aeration. In this process 10mg of algal biomass was homogenized with alkali (1ml of 10M aqueous KOH amended with 2.5% ascorbic acid) and heated (at 60°C) in a water bath for 10 min. |
