Comparison of cytochrome P450 CYP332A1 gene in resistant and susceptible strains of the cotton bollworm, Helicoverpa armigera (Lepidoptera: Noctuidae)

description The cotton bollworm, Helicoverpa armigera Hübner (Lepidoptera: Noctuidae), an important polyphagous pest, has shown different levels of resistance to various insecticides. Cytochrome P450 monooxygenases (P450s) are known to be closely related to insecticide resistance of H. armigera , but its internal mechanism is still not completely understood. In this study, CYP332A1 was cloned and characterized from fenvalerate-resistant (BJ) and susceptible (HDS) strains of H. armigera . The cDNA sequence of CYP332A1 contains an open reading frame (ORF) of 1509 bp encoding 502 amino acid residues. The expression level of CYP332A1 in BJ strain was significantly higher than that in HDS strain, and five amino acid mutations (F21C, G28R, K64Q, A290S and V477I) were observed in the sequence of BJ strain. The 5ʹ-flanking sequence of CYP332A1 contains the binding sites of putative transcription factors of Dr, Ubx, Cf2, caup, ara, Antp, ftz, eve and otp, and there were three large deletions of 386–392 bp, 547–554 bp and 965–971 bp in the 5ʹ-flanking sequence of CYP332A1 of BJ strain. The results suggested that CYP332A1 might play an important role in the resistance of H. armigera to fenvalerate, and the deletion of 5ʹ-flanking sequence of CYP332A1 might be one of the reasons for the development of resistance of H. armigera to fenvalerate.
