Summary:A protocol was developed to produce tetraploid and octoploid Haemanthus albiflos Jacq. (Amaryllidaceae) plants by treating with colchicine to diploid immature embryo-derived embryogenic calli. The calli were induced from immature embryos cultured on 3 g L −1 gellan gum-solidified Murashige and Skoog (MS) medium containing 5 mg L ‐1 each of picloram and 6-benzylaminopurine, and 30 g L −1 sucrose. Treatment with 0.2% (w/v) colchicine to the calli was performed with different durations of 24, 48, and 72 h. Tetraploids and octoploid plants were induced through somatic embryogenesis on a half-strength MS medium without plant growth regulators. Key message In diploid Haemanthus albiflos , immature embryo-derived embryogenic callus cultures were established. After colchicine treatment to the embryogenic callus cultures, tetraploid and octoploid plants were obtained simultaneously. Graphical abstract
