Summary:The assessment of genotoxicity or DNA damage in cancer cells is very crucial in evaluating the bioactive efficacy of potential anticancer compounds. Through single-cell gel electrophoresis (comet) assay, DNA damage can be visualized via staining with intercalating fluorescent dyes. This study ascertained the capacity of GelRed (GR) as an effective and equal alternative to the standard SYBR Green I (SG). Eight human cancer cell lines (MCF-7, H69PR, HCT-116, BxPC-3, HepG2, VCaP, HeLa, THP-1) were exposed to 5-fluorouracil and the levels of DNA damage were visualized via staining with GR and SG. Analyses of fluorescence photomicrographs via CometScore 2.0 revealed that there were no significant differences in the scoring parameter results afforded as tail length, % tail DNA, and tail moment among samples stained (p>0.05). The stability of the working solutions of the fluorescent dyes was also compared in a 30-day window period by performing comet assay on days 5, 10, 15, 20, 25, and 30, respectively.
