Summary:The study on stability of pollen sterility of three A 2 cytoplasm based male sterile lines (CORG 990047A, CORG 990052A and CORG 7A) showed that all the three male sterile lines had highly stable pollen sterility over 27 fortnight intervals. The various studies on pollen fertility, anther size and scanning electron microscopic image analysis revealed a clear insight on the anthers and pollen grain morphology of A and B lines. The nad7a_del marker distinguished the A and B lines of A 4 cytoplasm based male sterile lines by the allele size of 150 (10 bp deletion) and 160 bp, respectively. However, deletion of 10 bp was not observed in the A lines of A 2 cytoplasm (160 bp). Hence, this study proved that the nd7a_del marker can be used to differentiate A 2 and A 4 cytoplasm based male sterile lines. DNA fingerprinting study revealed that 12 SSR markers were polymorphic among the 47 markers used and two SSR markers viz., CcLG08_RFQI4 and CCM1277 were polymorphic and differentiated all the three-male sterile lines. These markers could be used in DNA fingerprinting of the A lines and to maintain their genetic purity.
