The various studies on pollen fertility, anther size and scanning electron microscopic image analysis revealed a clear insight on the anthers and pollen grain morphology of A and B lines. The

marker distinguished the A and B lines of A
4
cytoplasm based male sterile lines by the allele size of 150 (10 bp deletion) and 160 bp, respectively. However, deletion of 10 bp was not observed in the A lines of A
2
cytoplasm (160 bp). Hence, this study proved that the
nd7a_del
marker can be used to differentiate A
2
and A
4
cytoplasm based male sterile lines. DNA fingerprinting study revealed that 12 SSR markers were polymorphic among the 47 markers used and two SSR markers viz., CcLG08_RFQI4 and CCM1277 were polymorphic and differentiated all the three-male sterile lines. These markers could be used in DNA fingerprinting of the A lines and to maintain their genetic purity. |
