Summary: | Bacterial cultivation and inducible expression, followed by bacterial lysis and repeated rounds of precipitation/solubilization generated highly purified proteins. V96 and S48I48 were expressed in a bacterial system and were obtained as highly purified protein as a result of several precipitation/solubilization cycles. Specific phase transition were recorded for V96 at approximately 30°C, associated with aggregation and for S48I48 at approximately 25°C, associated with micelle formation. Generation of fusion proteins was achieved through a Ndel restriction site located at N-terminal of elastin-like polypeptides, which allowed insertion of EGFP and НА-ECTO sequences. |
