Summary:Highlights ► MARCKS is a frequent target of mutation in CRC with high-level microsatellite instability (MSI-H). ► Changing MARCKS content in CRC human and murine cell lines affects tumour-associated processes i.e. proliferation, migration and invasion. ► Loss of MARCKS protein hampers dynamic actin reorganization and interferes with the tumour-stromal cross talk between CRC and hHSCs. ► MARCKS might represent a suitable target to interfere and overcome the invasive behaviour of colon carcinoma cells at primary and distant sites.
