Summary: | Hexachlorobutadiene (HCBD) may be encountered as a by-product of hydrocarbon chlorination. The chronic toxicity and reproductive capacity of rats exposed to hcbd was studied. Dose levels of 2.0 or 20.0 mg/kg/day hcbd produced slight maternal toxicity, but did not affect reproductive parameters, such as percent pregnancy or neonatal survival and development. Decreased body weights were observed at the 20.0 mg/kg/day level. Chronic dietary studies showed multiple toxicological effects at the 20.0 mg/kg/day level, particularly of the kidney. Renal tubular adenomas and adenocarcinomas were observed. The low dose level of 0.2 mg/kg/day did not produce any apparent effects. Intermediate dose levels caused intermediate degrees of toxicity. A clear dose-response relationship is indicated for hcbd-induced toxicity. Renal neoplasms occurred only at dose levels higher than those causing obvious renal injury. |
