Summary: | The contaminant, 2-chlorodibenzofuran (2-MCDF), has been found in drinking water in Japan. In a study of teratogenicity, pregnant mice were exposed to various concentrations of 2-MCDF during organogenesis, from day 6 to day 15 of gestation. Liver weight relative to body weight increased in a dose-dependent manner in the pregnant females, demonstrating that the doses used were toxic to them. In the fetuses, several gross, visceral, and skeletal malformations were observed, but none were statistically significant. Some skeletal variations were observed, but only the incidence of hypoplastic supraoccipital bones was significant. The no-observable-effect levels for fetal and maternal mice were estimated to be 250 and 125 mg/kg/d, respectively. From this information, an allowable daily 2-MCDF intake for humans, and particularly, pregnant women, is 2.5 mg/d for a 50-kg woman, far higher than amounts that could reasonably be ingested from drinking water. |
