Lack of Estrogenic or (Anti-)antrogenic Effects of d-Phenothrin in the Uterotrophic and Hershberger Assays

d-Phenothrin is a type I pyrethroid pesticide with a low level of toxicity, but some in vitro studies have shown positive findings in the interaction of the pesticide with estrogen/androgen receptor, estrogen receptor agonist, and androgen receptor antagonist. description d-Phenothrin is a type I pyrethroid pesticide with a low level of toxicity, but some in vitro studies have shown positive findings in the interaction of the pesticide with estrogen/androgen receptor, estrogen receptor agonist, and androgen receptor antagonist. In this study, a 3-d uterotrophic assay using immature female rats and a 10-d Hershberger assay using castrated peripubertal male rats were performed to determine if d-phenothrin exhibited estrogenic or (anti-)androgenic activities in vivo. Ethynylestradiol, methyltestosterone, and p,p'-DDE were used as reference chemicals. In the uterotrophic assay, d-phenothrin caused no significant changes in clinical signs, body weight, food consumption, or kidney weights, but at a dose of 1000 mg/kg/d, liver weights were increased significantly, whereas uterine weights were not affected. In the Hershberger assay, liver weights were increased, but only at the 300 and 1000 mg/kg/d doses.
