Anaerobic induction of genes involved in the biosynthesis of amino acids and thiamine, in Escherichia coli

A series of fusions to lacZ was isolated by the insertional inactivation of genes involved in the production of acid from glucose. Description
Summary:A series of fusions to lacZ was isolated by the insertional inactivation of genes involved in the production of acid from glucose. Those which showed poor expression of lacZ aerobically, but increased production of β‐galactosidase anaerobically were further examined. Many of these were auxotrophic. Fusions to genes involved in the biosynthesis of leucine, isoleucine/valine, tyrosine and thiamine were induced anaerobically 2–10‐fold depending on the composition of the growth medium. Four fusion auxotrophs were mapped to known biosynthetic genes: WNK202 (thiamine; at 25 min near thiK), WNK203 (tyrosine; at 56.5 min in tyrA), WNK204 (leucine; at 2 min in leu) and WNK205 (isoleucine‐valine; at 84.5 min in the ilvG cluster). The leu and ilv auxotrophs WNK204 and WNK205 overproduced a common membrane protein when grown aerobically but not anaerobically.
