Summary:From a barley genomic library (partial Sau 3A digest of embryo DNA and lambda -Charon 35), the authors have isolated two different alpha -amylase genomic clones belonging to the high-pI family. Their identities were confirmed by hybridization to appropriate high-pI and low-pI alpha -amylase cDNA probes and by DNA sequencing. Comparison of these sequences with those of the alpha -amylase cDNA clones reveals that both of these high pI alpha -amylase genes contain three exons and two introns.
