Highlights • Prostate cancer cells over-express CPE-toxin receptors CLDN3 and CLDN4. • A protoxin was constructed that is PSA-protease-activated. • The protoxin was selectively cytotoxic to PSA-secreting cells. • The protoxin is a prototype for highly targeted prostate cancer therapy.
