Summary: | New macrocyclic lanthanide chelates that are potential contrast agents for magnetic resonance imaging have been synthesized. The ligands are derived from 1,4,7,10-tetraazacyclododecane-N,NN″,N″′-tetraacetic acid (DOTA) and feature one or two substituting groups directly grafted onto the tetraaza ring. The rigidity of the chelates as seen in the NMR spectra of paramagnetic species depends directly on the steric preferences of the macrocyclic unit. The solution structures of the rigid species are deduced from a quantitative analysis of the paramagnetic shifts. |
