Summary:Age-related muscle decline is associated with functional impairment of satellite cells (SCs). Conflicting data suggest dysregulation of cell-extrinsic or -intrinsic factors can independently contribute to such impairment. Here, we emphasize the importance of identifying nodes that integrate these factors into feed-forward circuits, which could provide targets for therapeutic intervention. Satellite cells generate and repair myofibers throughout the life of an organism, but their function declines with age. In this Forum, Sacco and Puri discuss how cell-intrinsic and -extrinsic factors are integrated to control rejuvenation of satellite cell function in aged animals.
